PDRN (polydeoxyribonucleotide) biostimulation uses concentrated DNA fragments extracted from wild salmon and injected via fine needle into the face, neck, décolletage or hands. The DNA extract is biocompatible with human tissue and works by binding to adenosine receptors on skin cells, stimulating fibroblast proliferation and collagen synthesis. Polynucleotides (PDRNs) are long-chain DNA fragments derived from the sperm cells of wild Pacific salmon, selected because salmon DNA shares a very high degree of structural homology with human DNA — making it biocompatible and non-allergenic for injection into human tissue. The PDRN molecule acts through two primary mechanisms in the skin. The first mechanism is purinergic receptor agonism: PDRN fragments are metabolised to adenosine, which binds to A2A receptors on fibroblasts, endothelial cells and macrophages. A2A receptor activation has a documented anti-inflammatory and pro-regenerative effect — reducing the inflammatory cytokine environment in damaged or aged skin and switching the cellular programme towards repair and remodelling. This is why PDRNs have also been studied in wound healing and diabetic ulcer management before their adoption into aesthetic medicine. The second mechanism is a direct mitogenic effect: polynucleotide fragments are incorporated into the PDRN salvage pathway, providing precursor material for DNA synthesis that supports accelerated cell turnover and proliferation in the treated zone. The combination of anti-inflammatory A2A agonism and direct DNA precursor provision produces a biostimulatory environment that drives fibroblasts to produce more collagen and elastin than they would under normal ageing conditions.
