In the tissue PDRN acts primarily by binding to P1 (adenosine A2A) receptors on fibroblasts and endothelial cells, triggering a downstream pathway that upregulates DNA synthesis and cell proliferation. Practically, this means fibroblasts begin producing more procollagen (primarily type I), elastin and hyaluronic acid — the three main structural components of youthful dermis. PDRN additionally promotes neovascularisation through VEGF upregulation and reduces the transcription of pro-inflammatory cytokines TNF-α and IL-6, making it particularly effective for chronically inflamed or sensitised skin.
